高效液相色谱法分析龙眼多糖的单糖组成

摘要


以新鲜龙眼果肉为原料,采用热水浸提法提取多糖,其提取工艺条件为:料液比(W∕V)1∶3,提取温度90℃,提取时间2h.2次重复实验的龙眼多糖得率为0.52%,总糖含量为20.23%,还原糖含量3.45%.多糖经硫酸水解后,通过PMP柱前衍生化处理后,以V(水)∶V(甲醇)=50∶50作流动相,1.0mL/min 的流速,检测波长254nm的色谱条件,利用高效液相色谱法进行分析.结果表明,龙眼多糖中存在L鼠李糖(LRha)、D葡萄糖(DGlc)、D半乳糖(DGal)3种单糖. This research used the fresh Longan as material and hot water extracting method to extract polysaccharides.The extraction conditions included material-to-solution ratio 1∶3,extraction temperature 90℃ extraction time 2h and experiments were repeated two times.The content of Longan polysaccharide,total sugar and reducing sugar was 0.52%,20.23% and 3.45%,respectively.After the polysaccharides was hydrolyed by sulfuric acid,the hydrolysate took PMP for Pre Column Derivatization,and the PMP derivatives were analysed by HPLC.With water:methanol(50∶50,V/V)as mobile phase,flow rate of 10mL/min,at the wavelength of 254nm analyse the PMP derivativesby HPLC analysis.The results show that Longan polysaccharide were composed of L-Rhamnose(L-Rha),D-Glucose(D-Glc),D-Galactose(D-Gal) three kinds of monosaccharides.
