剑叶龙血树树叶化学成分及其改善HepG-2细胞胰岛素抵抗的作用

摘要


采用硅胶、Sephadex LH-20和反相 RP C18等柱色谱技术, 从云南剑叶龙血树(Dracaena cochinchinensis)树叶甲醇和石油醚提取物中分离了7个化合物,包括3个黄酮类化合物,1个三萜和3个甾体化合物,经综合波谱分析分别鉴定为:4′,7-二羟基-3′-甲氧基-8-甲基黄烷(1),7-羟基-4′-甲氧基高异黄烷(2),2′,4,4′-三羟基查尔酮(3),(22E)-3β-acetoxystigmasta-5,22-diene(4),lanost-9-en-3β-ol(5),β-谷甾醇(6),胡萝卜苷(7).测定了化合物1~3改善HepG-2细胞对胰岛素抵抗的作用,化合物1~3能显著增加细胞的葡萄糖消耗量. This research focused on the chemical constituents isolated from the leaves of Dracaena cochinchinensis(Lour.)S.C.Chen.Seven compounds were obtained from the methanolic and petroleum ether extracts by various column chromatography on Si gel,Sephadex LH-20 and RP C-18,respectively.Their structures were identified as 4′,7-dihydroxy-3′-methoxy-8-methylflavan(1),7-hydroxy-3-(4′-methoxybenzyl)-4-chromanone(2),2′,4,4′-trihydroxychalcone(3),(22E)-3β-acetoxystigmasta-5,22-diene(4),lanost-9-en-3β-ol(5),β-sitosterol(6),daucosterol(7),based on the combined spectroscopic data and by comparison of these data with values from literatures.Compounds 1—3 were tested for their ameliorative effects on the insulin resistance in HepG-2 cell.Compounds 1—3 displayed relatively strong activities.
