Determination of the relative molecular mass of malanin with SDS-PAGE

蒜头果蛋白(malanin)是从广西、云南特有植物蒜头果中分离、纯化出的一种新的、具有抗肿瘤活性的植物蛋白质.采用变性十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)测定malanin的相对分子质量.结果表明,malanin是一种双链蛋白质,由A、B 2条肽链通过二硫键连接而成,其相对分子质量约为65.94×10^3.方法具有简单、快速、直观的特点. Malanin,a novel plant protein with anti-tumor activities,was isolated and purified from an endemic plant of malania oleifera in Guangxi and Yunnan provinces. The relative molecular mass of malanin was determined by the denaturing sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE). The result has showed that malanin is a protein with two chains,chain-A and chain-B,which are cross-linked by one or two disulfide bonds,and its relative molecular mass is about 65.94×10^3. SDS-PAGE gel electrophoresis is a simple,fast,direct-viewing method to determine the relative molecular mass of malanin.
